Indirect immunofluorescence assay for the detection of Pneumocystis carinii (now re-named Pneumocystis jirovecii) oocysts in human bronchoalveolar lavage and induced sputum.

Kit Components:
P.carinii monoclonal antibody (1ml), FITC-conjugated antibody (2ml), enzyme, dilute hydrochloric acid, enzyme diluent (3ml), specimen slides, mounting medium (5ml)
Assay Protocol:
1. Fix specimen to test slide with acetone
2. Add 20µl enzyme and incubate at 37°C for 30 minutes
3. Rinse and air dry
4. Add 15 µl of antibody and incubate at 37°C for 15 minutes
5. Rinse and air dry
6. Add 15µl FITC-conjugated antibody and incubate at 37°C for 5-15 minutes
7. Rinse and air dry
8. Add 1 drop of mounting medium and examine under fluorescence microscope
